Im wondering how to do this kind of thing:

how do you lumonate the brushes, also, how did he lumonate the render?

Most likely, you use the brush multiple times in the same spot to brighten it, instead of single clicking and then moving to a new brush and a new spot. Though thats only one step. Next would be Color Balance, as it would then illuminate it more than the darker brushes. Could also toy with Highlights if you want to.. though not many people use them.

Could go with a step of making one single layer for your bright spots, and then linking a color balance specifically to that layer to play with the settings for just those luminous spots.

Yup. When you are doing your black colored brushing, you want to switch to some white and put some white brushing in there, then you go to color balance and i believe in the section "highlights" is where you will get the best effect.
