After watching Channel 4's "What makes us Human?" http://www.channel4.com/science/microsites/W/what_makes_us_human/index.html

I'm interested in knowing what do you think makes us human?

Freedom is our defining attribute, I would say. It's a broad term, of course.

When I think about it there's no single defining thing to pick out about humanity that makes us what we are. Many animals have emotions and at least a rudimentary intelligence, freedom etc. It's the unique combination of these, and other, attributes that make up the range of humanity.
